If it's a registered right of way and the benefiting landowner behind you is willing to release it then it's very likely that you will need to both enter into a deed of release or similar. And then register that against both titles.
If anyone else enjoys the registered tight then they would need to be included also I suspect. And if your neighbour's title is mortgaged then the lender would need to also be involved/consent in most cases
Whilst you don't have to use a conveyancer we would always recommend that you do as they are familiar with the forms/process and legal requirements. And if mortgaged properties the lenders may insist you use one as well.
